This is a brief review of the leading anisotropic/diffusion-weighted MRI contrast agents. EMT, an analog of the natural peptide EGF, binds tightly to the EGF receptor (EGFR) more potently in vitro and in vivo than EGF. EMT induces cell death via a caspase-dependent pathway, but was relatively nontoxic to cells, organs, and mice due to its hydrophobic nature and fast clearance from plasma via the kidneys and liver with a half-life of approximately one day. It also effectively accumulated in tumors in both the subcutaneous and orthotopic models. The degree of target accumulation was dependent on EGFR expression levels and tumor type. Its toxicity was tested in Sprague-Dawley rats by injection into the tail vein for 13 days, in the dose of 40 mg/kg, with i.v. formulation. Although the rate of clearance of EMT was abnormally slow, plasma concentration did not decrease to zero within the duration of the experiment. The major effect was the induction of apoptosis in EGFR-positive tumor cells, which was easily detectable in tumor cells in vivo after one hour of its administration. No histological and biochemical damage was observed in organs and soft tissues, with the help of excluded-volume effects from the high tumor-to-blood volume. 7211a4ac4a
